Semax: The Brain Peptide You've Probably Never Heard Of
Semax is a synthetic heptapeptide (7 amino acids) originally developed at the Institute of Molecular Genetics in Russia. It's a modified fragment of ACTH (adrenocorticotropic hormone) ā specifically the 4-10 sequence ā engineered to retain neuroprotective and nootropic properties while eliminating the hormonal effects of full-length ACTH.
How Does Semax Work?
Semax's mechanism is multifaceted and still being fully characterised, but research has identified several key pathways:
BDNF Upregulation
Semax has been shown to increase Brain-Derived Neurotrophic Factor (BDNF) expression ā a protein critical for neuronal growth, survival, and synaptic plasticity. BDNF is often called "fertiliser for the brain."
Neurotransmitter Modulation
Research suggests Semax influences dopaminergic and serotonergic systems, potentially affecting focus, motivation, and mood regulation without the stimulant-like mechanisms of traditional nootropics.
Neuroprotection
Preclinical models show protective effects against oxidative stress and neuronal damage. Semax has been studied in ischaemic stroke models with positive outcomes in Russia.
Immune Modulation
Some research suggests Semax may influence immune function through its interaction with melanocortin receptors, though this is a less explored area of study.
Delivery Method
Unlike most peptides, Semax is typically administered intranasally (nasal spray) rather than by injection. The nasal route allows direct access to the brain via the olfactory pathway, bypassing the blood-brain barrier. This makes it one of the more user-friendly research peptides in terms of administration.
