IGF-1 LR3 vs HGH: Similar Goals, Very Different Science

⚔️ Comparison

HGH (Human Growth Hormone) and IGF-1 LR3 are often discussed together because they're part of the same biological axis — but they work at very different levels and have distinct research profiles. Understanding the distinction is important for anyone designing GH/IGF-1 pathway research.

Where They Sit in the Axis

HGH (Upstream)

191-amino acid protein. Released from the pituitary. Acts on the liver to produce IGF-1. Also has direct effects on fat metabolism, glucose regulation, and tissue growth. Works through the body's natural feedback loops.

IGF-1 LR3 (Downstream)

83-amino acid modified protein. Acts directly on IGF-1 receptors in tissues. Bypasses the pituitary entirely. No feedback regulation. Much more potent and longer-acting than native IGF-1 due to reduced IGFBP binding.

Key Differences

Mechanism

HGH works indirectly — it signals the liver to produce IGF-1, which then drives tissue growth. IGF-1 LR3 works directly on the end receptors, skipping the intermediary steps entirely.

Feedback Regulation

HGH maintains feedback loops — when GH and IGF-1 levels rise, the hypothalamus reduces GHRH output, keeping the system in balance. IGF-1 LR3 bypasses these controls entirely, which increases potency but also increases risk.

Selectivity

HGH has broad effects — fat metabolism, glucose regulation, tissue repair, immune function. IGF-1 LR3 is more narrowly focused on cell growth and proliferation via the PI3K/Akt/mTOR pathway.

Research Approach

For most GH axis research, secretagogues like ipamorelin offer the safest approach — they stimulate natural GH/IGF-1 production while preserving the body's feedback mechanisms. HGH introduces exogenous GH but maintains some feedback. IGF-1 LR3 is the most potent but also the least regulated option.

For GH axis research with maintained feedback control, we recommend ipamorelin as a starting point.
